目的:本实验以山楂叶总黄酮干预STZ-DN模型大鼠,观察山楂叶总黄酮对STZ-DN模型大鼠肾组织转化生长因子-β1(TGF-β1),骨形成蛋白-7(BMP-7)mRNA表达的影响。方法:采用链脲佐菌素(Streptozotocin,STZ)加高脂高糖喂养制作2型糖尿病大鼠模型。将Wistar大鼠48只随机分为正常对照组、STZ-DN模型对照组、山楂叶总黄酮组(FHL组)。结果:STZ-DN组大鼠血糖、BUN、BCr和尿蛋白明显增高;山楂叶总黄酮能够显著降低STZ-DN大鼠的血糖、尿蛋白并改善肾脏功能;STZ-DN组大鼠肾组织TGF-β1mRNA表达显著高于正常组,而FHL组则显著低于STZ-DN对照组;STZ-DN组大鼠肾组织BMP-7的mRNA表达较正常对照组显著减少,FHL组较STZ-DN组显著增多。结论:山楂叶总黄酮可通过上调BMP-7的表达,下调TGF-β1的表达从而对STZ-DN起治疗作用。
OBJECTIVE: To investigate the effect of hawthorn leaf flavonoids on the expression of transforming growth factor-β1 (TGF-β1) and bone morphogenetic protein-7 (BMP- 7) mRNA expression. Methods: Streptozotocin (STZ) plus high-fat and high-sucrose feeding rats model of type 2 diabetes. 48 Wistar rats were randomly divided into normal control group, STZ-DN model control group and hawthorn leaf flavonoids group (FHL group). Results: The levels of blood glucose, BUN, BCr and urine protein in STZ-DN group were significantly increased. The total flavonoids in hawthorn leaves could significantly decrease the blood glucose, urine protein and renal function in STZ-DN rats. The mRNA expression of BMP-7 in STZ-DN group was significantly lower than that in STZ-DN group, while the expression of BMP-7 mRNA in STZ-DN group was significantly lower than that of STZ-DN group in FHL group Significantly increased. Conclusion: The total flavonoids of hawthorn leaf can treat STZ-DN by up-regulating BMP-7 expression and down-regulating the expression of TGF-β1.
