目的 :建立一种外周血循环中肿瘤微转移检测的方法。方法 :2 0例CEA分泌性肿瘤患者及 8例健康对照 ,用逆转录PCR(ReverseTranscription PolymeraseChainReaction ,简称为RT PCR)方法检测血中的CEAmR NA ,同时用放免法检测血清CEA水平。结果 :2 0例患者CEAmRNA阳性率为 5 5 % ,而健康对照均为阴性。外周血CEAmRNA的存在与CEA分泌性肿瘤远处转移密切相关 ,而与CEA水平无明显关系。结论 :RT PCR是肿瘤微循环检测的灵敏方法 ,血中CEAmRNA阳性提示肿瘤的早期转移
Objective : To establish a method for detection of tumor micrometastasis in peripheral blood circulation. Methods : Twenty patients with CEA secretory tumors and eight healthy controls were tested for the CEAmR NA in the blood by Reverse Transcription Polymerase Chain Reaction (RT PCR). Serum CEA levels were also measured by radioimmunoassay. RESULTS: The positive rate of CEAmRNA was 55% in 20 patients, but it was negative in healthy controls. The presence of CEAmRNA in peripheral blood was closely related to the distant metastasis of CEA secreting tumors, but had no significant relationship with CEA levels. Conclusion: RT PCR is a sensitive method for detecting tumor microcirculation. Positive CEA mRNA in blood indicates early metastasis of tumor.
