目的:探讨高温诱导低分化鼻咽癌细胞系CNE-2Z凋亡的作用。方法:用激光共聚焦显微技术、核酸电泳、免疫组化等方法分析了42.5℃高温处理6、12、24h后鼻咽癌细胞的变化。结果:激光共焦显微镜观察可见高温处理6h肿瘤细胞出现空泡,有不同程度的固缩;12h染色质浓缩缘边化,呈“半月形”、细胞出芽,出现凋亡小体,24h后大部分细胞死亡。DNA琼脂糖凝胶电泳可见高温处理6h和12h后有断裂成小分子片段,呈典型梯状图谱;24h后为不规则的图谱。bcl-2免疫组化显示高温处理后的鼻咽癌细胞bcl-2蛋白表达逐渐减弱。结论:高温可诱导鼻咽癌细胞的凋亡,特别是42.5℃处理12h后明显。bcl-2蛋白与细胞凋亡之间关系密切。
Objective: To investigate the effect of high temperature on the apoptosis of poorly differentiated nasopharyngeal carcinoma cell line CNE-2Z. Methods: The changes of nasopharyngeal carcinoma cells after heat treatment at 42.5 ℃ for 6, 12 and 24 hours were analyzed by laser confocal microscopy, nucleic acid electrophoresis and immunohistochemistry. Results: Laser confocal microscopy showed that after 6 hours of high temperature treatment, the tumor cells showed vacuolization with different degrees of condensation. At 12th hour, the chromatin margin was marginalized and showed “half-moon shape” with sprouting and apoptotic bodies. After 24 hours, Some cells died. DNA agarose gel electrophoresis showed high temperature treatment 6h and 12h after breaking into small molecule fragments, showed a typical ladder map; irregular pattern after 24h. bcl-2 immunohistochemistry showed that bcl-2 protein expression in nasopharyngeal carcinoma cells gradually decreased after heat treatment. Conclusion: High temperature can induce the apoptosis of nasopharyngeal carcinoma cells, especially at 42.5 ℃ for 12h. Bcl-2 protein and apoptosis are closely related.
