目的:用高效液相色谱(HPLC)法与超高效液相色谱(UPLC)法对比测定千里香药材中九里香酮的含量。方法:分别建立了HPLC和UPLC测定千里香药材中九里香酮含量的方法。HPLC法为AKZONOBEL Kromasil 100-5C18(4.6mm×250mm,5μm)色谱柱,以乙腈-水(30∶70)为流动相,流速1mL.min-1,检测波长337nm,柱温30℃;UPLC法为WatersAcquityBEHC18(2.1mm×50mm,1.7μm)色谱柱,以乙腈-水(23∶77)为流动相,流速0.5mL.min-1,检测波长337nm,柱温30℃。结果:2种方法所得含量测定结果一致。HPLC法和UPLC法加样回收率的RSD分别是1.8%和1.7%。结论:HPLC和UPLC法测定千里香中九里香酮的含量,方法简便,结果准确可靠。UPLC较HPLC法更加简便迅速,既达到了分析的高通量,得到更高的分析灵敏度,又大大提高了分析速度,减少了有机溶剂的消耗。UPLC法能够成功替代HPLC法,为千里香药材质量控制提供新的检测方法。
OBJECTIVE: To compare and determine the content of glicinone in Chrysanthemum officinale with high performance liquid chromatography (HPLC) and ultra high performance liquid chromatography (UPLC). Methods: HPLC and UPLC were used to establish the method of determining the content of CM in Chinese herbs. The HPLC method was AKZONOBEL Kromasil 100-5C18 (4.6mm × 250mm, 5μm). The mobile phase was acetonitrile-water (30:70) with a flow rate of 1mL.min-1 and the detection wavelength was 337nm. As a Waters AccityBEHC18 (2.1 mm × 50 mm, 1.7 μm) column with acetonitrile-water (23:77) as the mobile phase at a flow rate of 0.5 mL · min-1 with a detection wavelength of 337 nm and a column temperature of 30 ° C. Results: The results obtained by the two methods were consistent. The RSDs of HPLC and UPLC were 1.8% and 1.7%, respectively. Conclusion: HPLC and UPLC method for the determination of the contents of nine miles fragrant incense, the method is simple, the results are accurate and reliable. Compared with HPLC, UPLC is more simple and rapid than HPLC, which not only achieves the high throughput of analysis but also achieves higher analytical sensitivity, greatly increases the analysis speed and reduces the consumption of organic solvents. The UPLC method can successfully replace the HPLC method and provide a new detection method for the quality control of Trichosanthes.
