目的比较不同产源的雷公藤多苷对大鼠睾丸组织病理改变及生精细胞凋亡的影响。方法 SD大鼠,随机分为4组,给予不同产源的雷公藤多苷灌胃,8周后取睾丸组织检测指标。光镜观察睾丸组织的形态学变化,原位末端标记法检测生精细胞凋亡率,免疫组化法检测凋亡相关蛋白Bax/Bcl-2的表达。结果睾丸生精细胞凋亡率和Bax/Bcl-2表达各组间有差异,其中Bax表达各给药组和空白组比较,差异有统计学意义(P<0.05),Bcl-2表达比较,各组间差异没有统计学意义(P>0.05)。结论促进Bax表达是雷公藤多苷生殖损伤的机理之一;三种不同产源的雷公藤多苷对雄性生殖的损伤作用有差异。
Objective To compare the effects of different sources of Tripterygium on the pathological changes of testis and the apoptosis of spermatogenic cells in rats. Methods SD rats were randomly divided into 4 groups, given different sources of tripterygium glycosides gavage, testicular tissue taken after 8 weeks index. Morphological changes of testis were observed with light microscope. Apoptosis rate of spermatogenic cells was detected by in situ end labeling. The expression of Bax / Bcl-2 protein was detected by immunohistochemistry. Results The apoptosis rate of spermatogenic cells and the expression of Bax / Bcl-2 in testis were different among groups. The expression of Bax in each administration group was significantly different from that in blank group (P <0.05) There was no significant difference among groups (P> 0.05). Conclusion The promotion of Bax expression is one of the mechanisms of reproductive injury of tripterygium wilfordii glycosides. Three different producing sources of tripterygium wilfordii glycosides have different effects on male reproductive injury.
