为观察乙肝病毒 H Bs Ag 基因疫苗免疫小鼠后的细胞及体液免疫应答,将该疫苗定向克隆于质粒pc D N A3 巨细胞病毒启动子下游,构建能在真核细胞内表达的重组基因疫苗pc D N A H Bs Ag 。将此疫苗通过多点肌肉注射免疫 Balb/ C 小鼠后第六周,实验组小鼠均出现抗- H Bs Ig G 阳性,其抗体水平明显高于对照组,同时 Th1 免疫应答相关的细胞因子 I L2 及γ干扰素水平也明显高于对照组,表明本文构建的基因疫苗pc D N A3 H Bs Ag 能诱导 Balb/ C 小鼠产生良好的细胞及体液免疫应答。
To observe the cellular and humoral immune response of mice immunized with HBsAg gene vaccine of hepatitis B virus, the vaccine was cloned into the downstream of the promoter of plasmid pc D N A3 cytomegalovirus to construct a recombinant gene vaccine capable of expression in eukaryotic cells pc D N A H Bs Ag. At the sixth week after immunization of Balb / C mice with this vaccine through multi-point intramuscular injection, all the mice in the experimental group showed positive anti-H Bs Ig G and their antibody levels were significantly higher than those in the control group. Th1 immune response-related cytokines I L 2 and γ interferon levels were significantly higher than the control group, indicating that the gene vaccine pcD N A3 H Bs Ag constructed in this paper can induce Balb / C mice to produce good cellular and humoral immune response.
