以兰科羊耳蒜菌根为试材,通过分离、纯化、培养其菌根真菌,在单因素试验基础上利用正交实验方法筛选温度、碳源、氮源和培养基pH值等培养条件来建立羊耳蒜菌根真菌培养体系。结果表明:羊耳蒜菌根真菌菌落白色或浅黄色,呈均匀辐射状向外生长;生长面有稀松或较丰富的气生菌丝,带有透明液滴。通过对菌落生长速率的测定与分析,4种因素的最佳综合水平为碳源为葡萄糖,浓度为2%;氮源为氯化铵,浓度为0.5%;pH为7,温度为25℃;菌落的生长速率随着天数的递增而以较快的速度下降,在第10天基本趋于一个恒定的值。
