目的探讨慢性期慢性粒细胞白血病(CML)患者骨髓间质干细胞(BMMSCs)体外生长特性和核转化因子κB(NF-κB)mRNA表达的变化及意义。方法选择5例慢性期CML患者为A组和6例健康人为B组,取骨髓单个核细胞行BMMSCs体外培养,观察BMMSCs体外生长形态并计数,绘制生长曲线。取第3、4代BMMSCs提取总RNA、逆转录RT-PCR检测NF-κB mRNA表达量。结果CML患者BMMSCs生长活性较对照组无明显差别,但其NF-κB mRNA表达量较对照组升高(P<0.05)。结论CML患者BMMSCs中NF-κB基因高表达影响CML发病。
Objective To investigate the in vitro growth characteristics of bone marrow mesenchymal stem cells (BMMSCs) and the expression of nuclear factor-κB (NF-κB) mRNA in patients with chronic myeloid leukemia (CML). METHODS: Five patients with chronic phase CML were selected as group A and six healthy individuals as group B. BMMSCs were obtained from bone marrow mononuclear cells in vitro. The growth morphology of BMMSCs was observed and counted, and the growth curve was drawn. Total RNA was extracted from the 3rd and 4th generation BMMSCs, and the expression of NF-κB mRNA was detected by RT-PCR. Results The growth activity of BMMSCs in CML patients was not significantly different from that in control group, but the expression of NF-κB mRNA in CML patients was higher than that in control group (P <0.05). Conclusion The high expression of NF-κB in BMMSCs of CML patients affects the pathogenesis of CML.
