目的探讨单羧酸转运蛋白-1(MCT1)、CD147、CD44 mRNA在非小细胞肺癌(NSCLC)中的表达及其在NSCLC发生、发展和预后中的意义。方法取经病理确诊的56例NSCLC患者的癌及癌旁组织(距肿瘤边缘>5 cm)、20例肺良性病变、7例正常肺组织组织标本,采用实时荧光定量RT-PCR检测MCT1、CD147、CD44 mRNA表达,并分析癌组织中三者表达的相关性及与临床病理特征的关系。结果 (1)MCT1、CD147、CD44 mRNA在肺癌组织中的表达量高于癌旁组织、正常肺组织和肺良性病变组织,差异均有统计学意义(P均<0.05);(2)癌组织中MCT1、CD147、CD44 mRNA三者表达具有相关性(P<0.05);(3)MCT1、CD147、CD44 mRNA的表达与NSCLC患者的性别、年龄、是否吸烟、肿瘤大小、pTNM分期均无关,但与肿瘤的组织类型(P均<0.05)、淋巴结转移(P均<0.05)有关;MCT1、CD44 mRNA的表达还与肿瘤分化程度有关(P均<0.05)。结论在NSCLC患者组织中MCT1、CD147、CD44 mRNA表达上调,它们可能参与肿瘤的发生、发展,有望成为NSCLC诊断和预后评价的指标。
Objective To investigate the expression of monocarboxylate transporter 1 (MCT1), CD147 and CD44 mRNA in non-small cell lung cancer (NSCLC) and their significance in the development, progression and prognosis of NSCLC. Methods Totally 56 cases of NSCLC patients were examined by real-time fluorescence quantitative RT-PCR to detect the expression of MCT1, CD147, CD147 in cancer and adjacent tissues (> 5 cm from tumor margin), 20 cases of benign lung disease and 7 cases of normal lung tissue. CD44 mRNA expression, and analysis of the correlation between the expression of three in cancer tissue and clinicopathological features. Results (1) The expression of MCT1, CD147 and CD44 mRNA in lung cancer tissues was significantly higher than that in paracancer tissues, normal lung tissues and benign lung lesions (all P <0.05). (2) (P <0.05). (3) The expressions of MCT1, CD147 and CD44 mRNA had no correlation with the gender, age, smoking status, tumor size and pTNM stage in patients with NSCLC, but there was no correlation between the expression of MCT1, CD147 and CD44 (P <0.05), and lymph node metastasis (all P <0.05). The expression of MCT1 and CD44 mRNA also correlated with the degree of tumor differentiation (all P <0.05). Conclusion The expressions of MCT1, CD147 and CD44 mRNA in NSCLC patients are up-regulated, which may be involved in the occurrence and development of tumors. It is expected to be an index for the diagnosis and prognosis evaluation of NSCLC.
