目的探讨碘化钾(KI)对人皮肤成纤维细胞(HSF)胶原合成的影响。方法将体外培养的HSF分为5组,KI组分别加入3、30、300 mg/L的KI,bFGF组加入3 mg/L bFGF,对照组加入同等剂量的无菌生理盐水,作用96 h,倒置显微镜下观察各组HSF形态学变化,采用Real-Time PCR法检测Ⅰ型胶原(COL1)、Ⅲ型胶原(COL3)mRNA,采用3H-脯氨酸比色法测定成纤维细胞胶原的合成。结果与对照组比较,3 mg/L KI组COL3 mRNA表达升高,bFGF组COL1、COL3 mRNA表达升高(P均<0.05);与对照组比较,3、30 mg/L KI组及bFGF组培养液中胶原合成增高,3、30、300 mg/L KI组及bFGF组细胞内胶原合成增高(P均<0.05)。结论适量浓度的KI可使人皮肤成纤维细胞合成和分泌胶原增加。
Objective To investigate the effect of potassium iodide (KI) on collagen synthesis of human dermal fibroblasts (HSF). Methods The HSF cultured in vitro were divided into five groups. The KI groups were treated with 3, 30, and 300 mg / L KI respectively, and the bFGF group with 3 mg / L bFGF. The control group was given the same amount of sterile saline for 96 hours, The morphological changes of HSF in each group were observed under inverted microscope. The mRNA expression of COL1 and COL3 was detected by Real-Time PCR. The synthesis of fibroblasts was measured by 3H-proline colorimetric assay. Results Compared with the control group, COL3 mRNA expression was increased in 3 mg / L KI group and COL1 and COL3 mRNA expressions in bFGF group were increased (all P <0.05). Compared with the control group, 3,30 mg / L KI group and bFGF group Collagen synthesis increased in the culture medium, and collagen synthesis was increased in 3, 30, 300 mg / L KI group and bFGF group (all P <0.05). Conclusion KI at appropriate concentration can increase the synthesis and secretion of collagen in human dermal fibroblasts.
