在免疫酶组织化学技术中,内源性过氧化物酶的干扰是影响结果判断的一个重要因素。迄今为止文献报道的各种抑制内源性过氧化物酶的方法都不理想,其缺点不是内源性过氧化物酶未能完全抑制,就是损伤了抗原的反应活性。作者研究出一种新的方法,既不损伤抗原活性又能完全消除内源性过氧化物酶。方法:二硝基氟苯等诱发小鼠产生炎症,取耳组织等作冰冻切片,丙酮固定。用6种抗鼠的单克隆抗体(抗Lyt1、Lyt2和L3T4,抗Ia,BM8和7D10)作组化染色。
In the immunoenzymatic histochemical technique, the interference of endogenous peroxidase is an important factor affecting the judgment of the result. So far, various methods for inhibiting endogenous peroxidase have not been reported in the literature. The disadvantage is not that endogenous peroxidase can not completely inhibit or impair the antigen’s reactivity. The authors have developed a new approach that does not damage antigen activity but completely eliminates endogenous peroxidases. Methods: Inflammation was induced in mice such as dinitrofluorobenzene, and the ear tissues were frozen and fixed with acetone. Six anti-mouse monoclonal antibodies (anti-Lyt1, Lyt2 and L3T4, anti-Ia, BM8 and 7D10) were used for histochemical staining.
