目的探讨牛蒡子萃取物对血瘀证动物模型血液流变学的影响。方法 Wistar大鼠60只,按体重随机分为假手术组、模型组、阳性组、牛蒡子提取物高、中、低剂量组,每组10只,除假手术组外,每组分别给予相应药物7 d,末次给药30 min后,各组动物皮下注射盐酸肾上腺素注射液,间隔6 h后同上述方法再注射一次,两次注射盐酸肾上腺素注射液之间将动物置于2℃冰水浴中游泳5 min,制备血瘀动物模型,假手术组不造模。检测大鼠血液流变学指标、凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、纤维蛋白原(FIB)、血小板聚集率(PA)。结果与模型组比较,阳性组、高、中剂量组的全血黏度有所降低(P<0.05);阳性组,高、中、低剂量组的PT、APTT升高,FIB含量有所降低(P<0.05);阳性组PA有显著性降低(P<0.01),高剂量与中剂量组有差异(P<0.05),与低剂量组无差异。结论牛蒡子萃取物具有降低血瘀证大鼠全血黏度、改善血液流变学指标的作用;牛蒡子萃取物能够改善凝血功能,降低血小板聚集。
Objective To investigate the effects of Arctium lappa L. extract on hemorheology in animal model of blood stasis. Methods Sixty Wistar rats were randomly divided into sham operation group, model group, positive group and high, medium and low dose group of Arctium lappa extract, with 10 rats in each group. Except sham group, Drugs for 7 days, the last administration of 30 min, each group of animals injected subcutaneously with epinephrine hydrochloride injection 6 hours after the above method with a second injection, the injection of two epinephrine hydrochloride between the animals placed in 2 ℃ ice Swimming in water bath for 5 min, preparation of blood stasis animal model, sham operation group not modeling. The indexes of hemorheology, prothrombin time (PT), activated partial thromboplastin time (APTT), fibrinogen (FIB) and platelet aggregation rate (PA) were detected. Results Compared with the model group, the whole blood viscosity of the positive group, high dose group and middle dose group decreased (P <0.05), while the PT, APTT, FIB content of the positive, P <0.05). In the positive group, PA was significantly decreased (P <0.01). There was difference between the high dose and middle dose groups (P <0.05), but no difference with the low dose group. Conclusion Fructus Arctii extract can reduce whole blood viscosity of blood stasis rats and improve the indexes of hemorheology. Fructus Arctii extract can improve blood coagulation and reduce platelet aggregation.
