目的:探讨湖北海棠总黄酮对去势大鼠骨质疏松的影响。方法:取三月龄SD雌性大鼠经去势手术或假手术后,喂以去大豆特殊饲料7周以诱导骨丢失。将去势大鼠分为模型组,湖北海棠总黄酮组(23.3、46.6、93.3mg/kg),己烯雌酚组,连续给药12周。检测大鼠血清中骨钙素,雌二醇的含量,尿液中脱氧吡啶啉的含量,血清和尿液中Ca、P含量,观察大鼠腰椎骨末端骨小梁的变化情况并计算骨小梁面积百分比。结果:湖北海棠总黄酮降低了大鼠血清中骨钙素以及尿液中脱氧吡啶啉的含量,与模型组相比,46.6mg/kg和93.3mg/kg剂量组差异有显著性;湖北海棠总黄酮提高了大鼠血清中雌二醇含量,但与模型组相比,无统计学意义;湖北海棠总黄酮提高了大鼠血清钙含量,降低大鼠尿液中钙、磷的含量,93.3mg/kg组作用最显著;骨小梁的形态学观察及数据分析显示,与模型组相比,湖北海棠总黄酮可以提高骨小梁的厚度和面积,其46.6 mg/kg和93.3mg/kg明显地提高了骨小梁面积百分比。结论:湖北海棠总黄酮可以降低去势大鼠骨转换率和骨吸收程度,对绝经后骨质疏松具有防治作用,其作用机制可能与湖北海棠总黄酮中根皮苷等黄酮成分具有雌激素类似结构,能与雌激素受体结合,发挥雌激素样作用有关。
Objective: To investigate the effects of Begonia officinalis flavonoids on osteoporosis in ovariectomized rats. METHODS: Three-month-old SD female rats were either castrated or sham-operated and fed with special soy-based diet for 7 weeks to induce bone loss. The castrated rats were divided into model group, Hubei Begonia total flavonoids group (23.3,46.6,93.3mg / kg), diethylstilbestrol group, continuous administration for 12 weeks. Serum levels of osteocalcin, estradiol, content of deoxypyridines in urine, content of Ca and P in serum and urine were measured. The changes of trabecular bone in the lumbar vertebrae were observed, Beam area percentage. Results: The total flavonoids of Begonia officinalis reduced the content of serum osteocalcin and deoxypyridinine in urine, compared with the model group, the differences were significant between the 46.6mg / kg and 93.3mg / kg dose groups; Flavonoids increased the content of estradiol in rat serum, but there was no significant difference compared with model group. The total flavonoids of Begonia officinarum increased serum calcium and the content of calcium and phosphorus in rat urine, 93.3mg / kg group was the most significant; trabecular morphology and data analysis showed that compared with the model group, the total flavonoids of Begonia can increase the thickness and area of trabecular bone, 46.6 mg / kg and 93.3 mg / kg significantly Increased trabecular area percentage. CONCLUSION: The total flavonoids from Begonia caulis can reduce the rate of bone turnover and bone resorption in ovariectomized rats, and have a preventive and therapeutic effect on postmenopausal osteoporosis. The mechanism may be related to the similar estrogen-like structure of flavonoids in the total flavonoids of Begonia cauliflower , With estrogen receptor binding, play an estrogen-like role.
