目的了解江西省流行的呼吸道合胞病毒(RSV)的G蛋白基因特征。方法收集2005年-2007年流感样病例标本,进行RSV分离;对病毒G蛋白进行全基因序列测定,并与国际标准株进行比较,对G蛋白潜在的O-糖基化位点进行分析。结果从4291份标本中分离到6株RSV。各分离株与A、B型标准株的氨基酸同源性分别为85.6%~87.6%和75.9%~76.7%;其3’端基因高变区核苷酸序列与RSV GA2亚型的同源性最高。分离株在人群免疫选择压力位点111、132、226、262、274、290、297位氨基酸上发生了变异,与A2株比较,众多的潜在的O-糖基化位点发生了改变。结论 2005年-2007年江西省RSV流行株属于GA2亚型,在G基因的人群免疫选择压力位点和糖基化位点有变异。
Objective To understand the G protein gene characteristics of the respiratory syncytial virus (RSV) in Jiangxi Province. Methods Samples of influenza-like illness from 2005 to 2007 were collected for RSV isolation. The whole G-protein sequence of the virus was determined and compared with the international standard strains to analyze the potential O-glycosylation sites of G protein. Results Six strains of RSV were isolated from 4291 specimens. The amino acid homologies between the isolates and the standard strains A and B were 85.6% -87.6% and 75.9% -76.7%, respectively. The homology of the 3’-terminal hypervariable region nucleotide with the RSV GA2 subtype highest. The isolates mutated at amino acids 111, 132, 226, 262, 274, 290 and 297 in the immunocompetent stress site of the human population. Compared with strain A2, many potential O-glycosylation sites were altered. Conclusion From 2005 to 2007, the epidemic strains of RSV in Jiangxi Province belonged to the subtype GA2, and there were variations in the pressure-selective and glycosylation sites of G-selectin population.
