目的研究不同洗脱液对环氧乙烷生物指示剂的菌量及抗力测定结果的影响。方法通过细菌定量检测方法,比较不同洗脱液对生物指示剂上活菌洗脱效果及其对生物指示剂抗力的影响。结果 6种洗脱液与5种稀释液对同一批次的生物指示剂载体上细菌洗脱后,细菌回收效果最好的是用5 000 mg/L吐温-80的营养肉汤洗脱后,用浓度为5 000 mg/L吐温-80的PBS,回收菌数为7.03×106cfu/片。回收效果最差的是无菌生理盐水,平均回收菌数为3.78×106cfu/片。回收菌数数值越大D值越小,ST值相应变大和KT值变小。结论洗脱效果最好的洗脱液和相应稀释液是吐温-80营养肉汤与吐温-80 PBS,生物指示剂回收菌数越高,其D值更接近真值。
Aim To study the effect of different eluents on the determination of the biological quantity and the resistance of ethylene oxide biological indicators. Methods The bacterial elution method was used to compare the elution effects of different eluents on the viability of biological indicators and their effects on the biomarker resistance. Results After the bacteria were eluted from the same batch of biological indicator carrier with 6 kinds of elution liquid and 5 kinds of diluent, the best bacteria recovery effect was after the elution of nutrient broth with 5 000 mg / L Tween-80 , With a concentration of 5000 mg / L Tween-80 in PBS, the number of bacteria was 7.03 × 106cfu / piece. The worst recovery is sterile saline, the average number of recovered bacteria 3.78 × 106cfu / piece. The larger the number of recovered bacteria, the smaller the value of D, the larger the value of ST and the smaller the value of KT. Conclusion The best eluant and the corresponding diluent were Tween-80 nutrient broth and Tween-80 PBS. The higher the biological indicator was, the closer the D value was to the true value.
