我们采用氚-胸腺嘧啶核苷(~3H-TdR)和碳~■尿嘧啶核苷(~(14)C-UR)双标记淋巴细胞转化法,观察白血病患者淋巴细胞转化过程中两种核酸的合成与代谢情况。现报道如下:一、~3H-TdR、~(14)C-UR 双标记掺入试验:每只培养管装2ml 含PHA16μg 的772培养液(均为上海东风试剂厂产品),调pH 至7.5左右,加注0.2ml 肝素抗凝全血,37℃孵育70h 后,加入~3H-TdR1.13μCi(放射性比度14Ci/mM,上海原子核所产品)和~(14)C-UR0.2μCi(放射性比度53mCi/mM,中国医科院简阳分院产品),继续孵育6h。收获时每管加蒸馏水5ml,破坏红细胞,移至49型玻璃纤维滤膜上抽滤;再用生理盐
