目的观察赤霉素(gibberellins,GA)的添加量对球等鞭金藻3011(Isochrysis galbana 3011)生物量、细胞形态与不同生长时期群体细胞活性的影响。方法取处于指数生长期的球等鞭金藻3011藻液,分别添加终浓度为0(溶剂对照)、0.001、0.003、0.005、0.007、0.01、0.03、0.05、0.07、0.1 mg/L的赤霉素溶液,测定球等鞭金藻3011的密度和细胞形态;采用流式细胞技术对指数生长期(第8天)、相对生长下降期(第13天)和静止期(第18天)的球等鞭金藻3011群体细胞活性进行测定。结果添加0.005 mg/L赤霉素可使球等鞭金藻3011的生物量达到最高(1.76×108/ml);且与溶剂对照组比较,球等鞭金藻3011细胞面积增大,亮度提高(P<0.05或P<0.01)。0.07 mg/L赤霉素可使相对生长下降期群体细胞的活性最高(99.73%),引起指数生长期延长。结论 0.005 mg/L赤霉素可促进球等鞭金藻3011生物量和细胞形态指数的增加;0.07 mg/L赤霉素可提高群体细胞活性,延长了指数生长期。
Objective To observe the effect of gibberellins (GA) on biomass, cell morphology and cell population in different growth stages of Isochrysis galbana 3011. Methods The algae of Isochrysis galbana 3011 in the exponential phase of growth were harvested, and the final concentrations of 0 (solvent control), 0.001,0.003,0.005,0.007,0.01,0.03,0.05,0.07 and 0.1 mg / The density and cell morphology of Isochrysis galbana 3011 were determined by flow cytometry. The effects of flow cytometry on the growth of exponential growth phase (day 8), relative growth decline phase (day 13) and quiescence (day 18) Isochrysis galbana 3011 population cell viability was measured. Results The biomass of Isochrysis galbana 3011 reached the highest level (1.76 × 108 / ml) with addition of 0.005 mg / L gibberellin. Compared with the solvent control group, the area of Isochrysis galbana 3011 cells increased and the brightness increased (P <0.05 or P <0.01). The concentration of gibberellin (0.07 mg / L) could increase the cell population in the relative growth phase (99.73%) and prolong the exponential growth phase. Conclusions GA3 0.01 mg / L could promote the biomass and cell morphology index of Isochrysis galbana 3011. GA 0.07 mg / L could increase the cell viability and prolong the exponential growth phase.
