采用C(18)小柱净化尿样,富集尿中被测物丙烯酰胺代谢物──巯基尿酸(乙酰半胱氨酸-S-丙酰胺)。处理过的尿样用Ellman's反应分光光度法测定。用本方法做游离巯基测定的批内变异系数为0.14%~1.6%(n=6),总巯基测定的批内变异系数为1.0%~5.2%(n=6),方法的加标回收率为82.7%~84.4%。检出下限:游离巯基尿酸为0.008mmol/L,总巯基尿酸为0.013mmol/L。本方法测定了9例非接触者尿样,巯基尿酸浓度范围0.056~0.086mmol/L,41例接触丙烯酰胺工人的尿,巯基尿酸浓度范围0.061~0.374mmol/L。
C (18) cartridge was used to purify the urine sample and enrich the metabolite of acrylamide, namely thiol uric acid (acetylcysteine-S-propionamide) in the urine. Treated urine samples were measured by Ellman’s reaction spectrophotometry. The intra-assay coefficient of variation (ICC) of this method was 0.14% -1.6% (n = 6) and the intra-assay coefficient of variation of total sulfhydryl was 1.0% -5.2% (n = 6 ), The recovery rate of the method was 82.7% ~ 84.4%. Lower limit of detection: Free thiol uric acid 0.008mmol / L, total thiourea uric acid 0.013mmol / L. The urine samples of 9 non-contact persons were measured by this method. The concentrations of thiourea and uric acid were 0.056-0.086 mmol / L, while the urine samples of 41 patients exposed to acrylamide were 0.061-0.374 mmol / L.
