随着超低温技术的日趋成熟,其应用领域不断拓展,特别是在造血干细胞移植过程中,该技术起着非常关键的作用,而冷冻对免疫细胞影响的大小关系到机体免疫状况.为了探讨超低温冷冻对人单个核细胞(PBMC)中不同淋巴细胞亚群活率的影响是否相同,本研究对23例行自体外周血干细胞移植治疗的实体瘤患者PBMC中各淋巴细胞亚群(CL4~+,CD8~+,CD45~+,CD56~+)冻存前及复苏后的活率进行了检测.我们用23例实体瘤患者中,乳腺癌7例,恶性淋巴瘤10例,卵巢癌2例,生殖细胞肿瘤1例,肺癌1例,多发性骨髓瘤1例.年龄30~55岁.使用美国百特公司血细胞分离机(CS-3000Plus)采集PBMC.每份样本的冻存体积为60ml.冷冻保护液为70%Tc-199,20%DMSO,10%AB血清.应用程控降温系统(美国Froma公司)使样本温度降至-
With the development of ultra-low temperature technology, its application fields are expanding, especially in the process of hematopoietic stem cell transplantation, which plays a crucial role in the immune system, and the impact of freezing on the immune cells is related to the immune status.In order to investigate the effects of cryopreservation (PBMC) in different lymphocyte subsets of the same, the study of 23 patients with autologous peripheral blood stem cell transplantation in patients with solid tumor PBMC lymphocyte subsets (CL4 ~ +, CD8 ~ +, CD45 ~ +, CD56 ~ +) were detected before and after resuscitation.We used 23 cases of solid tumor patients, 7 cases of breast cancer, 10 cases of malignant lymphoma, 2 cases of ovarian cancer, reproductive 1 case of cell tumor, 1 case of lung cancer and 1 case of multiple myeloma, aged from 30 to 55. The PBMCs were collected using a CS-3000Plus, a cryopreservation volume of 60 ml for each sample. The fluid was 70% Tc-199, 20% DMSO, 10% AB Serum.
