目的提高实验室分子检测水平,强化实验室人员的质量意识,提高血液检测的安全性和有效性。方法2015年10月至11月参加由德国INSTAND组织的国际医学实验室室间质量评估项目,对发放的未知DNA标本,用PCR-SSP的方法对ABO、Rh、MNS、Kell、Kidd和Duffy等主要血型抗原系统进行基因分型检测。结果对发放的4个DNA标本分别进行抗原的基因分型,结果依次为:61号,B/O1,KEL2/KEL2,JKA/JKB,FYA/FYA,MNs,Ccee,RHD Gen/gene(normal);62号,O1/O1,KEL2/KEL2,JKA/JKB,FYA/FYB,MNs,Ccee,RHD Gen/gene(normal);63号,O1/O1,KEL2/KEL2,JKA/JKB,FYA/FYA,NNs,CCee,RHD Gen/gene(normal);64号,O1/O1,KEL2/KEL2,JKA/JKB,FYB/FYB,MNs,Cc Ee,RHD Gen/gene(normal)。结论鉴定结果与质评反馈结果完全一致。该项目提高了输血医学实验室分子检测的水平,验证了实验室体系的稳定性、保证临床输血检测的安全性和有效性。
Objective To improve the level of laboratory molecular tests, enhance the quality awareness of laboratory personnel and improve the safety and effectiveness of blood tests. Methods From October to November 2015, we participated in the International Medical Laboratory Inter-laboratory Quality Assessment Program organized by INSTAND in Germany. We used PCR-SSP to analyze the unknown DNA samples of ABO, Rh, MNS, Kell, Kidd and Duffy The major blood group antigen system for genotyping. Results The genotypes of antigens in four DNA samples were sequenced. The results were as follows: 61, B / O1, KEL2 / KEL2, JKA / JKB, FYA / FYA, MNs, Ccee, RHD Gen / ; No. 62, O1 / O1, KEL2 / KEL2, JKA / JKB, FYA / FYB, MNs, Ccee, RHD Gen / gene , NNs, CCee, RHD Gen / gene (normal); 64, O1 / O1, KEL2 / KEL2, JKA / JKB, FYB / FYB, MNs, Cc Ee, RHD Gen / gene (normal). Conclusion The result of the appraisal is exactly the same as that of the quality appraisal feedback. The project has raised the level of molecular testing in transfusion medicine laboratories, validated the stability of the laboratory system and ensured the safety and effectiveness of clinical transfusion tests.
