目的研究痛风饮中有效成分橙皮苷、儿茶素和芒果苷在大鼠体内药代动力学。方法采用HPLC法,测定SD大鼠灌胃给予痛风饮后不同时间橙皮苷、儿茶素和芒果苷的血药浓度,采用DAS 2.0软件计算3种成分的药物代谢动力学参数。结果橙皮苷在0.13~2.67μg·ml~(-1)呈线性关系,药动学参数:C_(max)为(22.40±0.22)μg·ml~(-1),T_(max)为1 h,AUC_(0-t)为(155.11±1.36)mg·L~(-1)·h;芒果苷在0.17~3.44μg·ml~(-1)呈线性关系,药动学参数:C_(max)为(12.13±0.18)μg·ml~(-1),T_(max)为2 h,AUC_(0-t)为(181.84±1.51)mg·L~(-1)·h;儿茶素在0.08~1.65μg·ml~(-1)呈线性关系,药动学参数:C_(max)为(18.00±0.26)μg·ml~(-1),T_(max)为10 h,AUC_(0-t)为(248.66±1.55)mg·L~(-1)·h。结论大鼠单次灌胃痛风饮后,橙皮苷、儿茶素和芒果苷体内过程均呈二室模型。
Aim To study the pharmacokinetics of hesperidin, catechin and mangiferin in gout drink in rats. Methods HPLC method was used to determine the plasma concentrations of hesperidin, catechin and mangiferin at different times after oral administration of Tongfeng drink in SD rats. The pharmacokinetic parameters of three components were calculated by DAS 2.0 software. Results Hesperidin showed a linear relationship in the range of 0.13-2.66 μg · ml -1. The pharmacokinetic parameters were as follows: C max of 22.40 ± 0.22 μg · ml -1 and T max of 1 h, AUC_ (0-t) was (155.11 ± 1.36) mg · L -1 · h, and the linear relationship was found between 0.17 and 3.44 μg · ml -1. The pharmacokinetic parameters were C_ ( max was (12.13 ± 0.18) μg · ml -1, T max was 2 h, and AUC 0-t was (181.84 ± 1.51) mg · L -1 · h. The linear range was 0.08-1.65μg · ml -1. The pharmacokinetic parameters were as follows: C max 18.00 ± 0.26 μg · ml -1, T max 10 h, (0-t) was (248.66 ± 1.55) mg · L -1 · h. Conclusion After a single oral administration of Gout in rats, hesperidin, catechin and mangiferin in vivo showed a two-compartment model.
