水稻初级三体在水稻遗传育种中起着重要的作用。其归属鉴定通常用 Shastry(1960 )的粗线期核型分析方法 ,但这种方法在实际操作中有一定的难度 ,不易得到分散好的粗线期染色体分裂相。本研究首次尝试利用三体株和正常株同源染色体间 DNA含量的差异 ,用分子标记的手段将量的差异转为自显影带的差异原理去鉴定三体 ,发现此鉴定结果与粗线期核型分析结果一致 ,而此方法在实验技术上可靠性高 ,快捷 ,因此是一种新的有效鉴定初级三体的方法
Rice trisomic body plays an important role in rice genetic breeding. Attribution identification is usually based on the pachytene karyotype analysis method of Shastry (1960). However, this method has some difficulties in practical operation and it is not easy to obtain a well-dispersed pachytene split phase. This study, for the first time, attempted to use the difference of DNA content between homologous chromosomes of the three-body and normal strains and to use the molecular marker method to convert the difference of the amount into the self-development zone to identify the trisomies. The results of this identification were consistent with those of the pachytene Karyotype analysis results are consistent, and this method is highly reliable in experimental techniques, fast, and therefore a new method for the effective identification of primary trisomy
