目的供体抗原特异性的CD4+CD25+Treg体内应用后对同种异系肾移植大鼠受体B细胞功能的影响。方法以F344大鼠为供体、Lewis大鼠为受体建立大鼠肾移植模型,于术前1天、术后2周分别将供体抗原特异性的CD4+CD25+Treg(1×106)于尾静脉注入受体体内,术后1月抽取大鼠外周血,分离B细胞,检测其在抗IgM抗体刺激下的增殖情况,检测大鼠外周血IgG、IgA水平,以判断应用CD4+CD25+Treg后大鼠体内B细胞功能状态。实验分组:实验组术前、术后2周使用CD4+CD25+Treg(n=8),阳性对照组术前、术后未使用CD4+CD25+Treg(n=8),空白对照组未移植大鼠(n=8)。结果实验证实实验组B细胞增殖水平最低、空白对照组其次,阳性对照组最高,三者均有显著差异(P<0.05),外周血IgG、IgA水平检测证实,实验组与空白对照组无显著差异(P>0.05),二者明显低于阳性对照组(P<0.01)。结论经尾静脉注入CD4+CD25+Treg后肾移植受体体内B细胞的功能可明显受到抑制,CD4+CD25+Treg可通过对B细胞的这一作用调节受体免疫状态,从而达到控制排斥反应,诱导免疫耐受的作用。
Objective To investigate the effect of donor antigen-specific CD4 + CD25 + Treg on the function of recipient B cells in allogeneic renal allograft rats. Methods F344 rats were used as donors and Lewis rats were used as recipients to establish rat kidney transplantation model. The donor antigen - specific CD4 + CD25 + Treg (1 × 106) Peripheral blood was collected from the tail vein and the peripheral blood was collected from January 1, and B cells were isolated. Proliferation of the cells was detected by anti-IgM antibody. The levels of IgG and IgA in peripheral blood of rats were measured to determine whether CD4 + CD25 + Treg in rats after B cell function status. The experimental group: CD4 + CD25 + Treg (n = 8) was used preoperatively and 2 weeks after operation in the experimental group, CD4 + CD25 + Treg was not preoperatively and postoperatively in the positive control group, but not in the blank control group Rat (n = 8). Results The results showed that the proliferation of B cells in the experimental group was the lowest, the highest in the blank control group and the highest in the positive control group (P <0.05). The levels of IgG and IgA in the peripheral blood of the experimental group were significantly lower than those of the blank control group (P> 0.05), which were significantly lower than those in the positive control group (P <0.01). Conclusion The function of B cells in recipients of renal allograft via tail vein injection of CD4 + CD25 + Treg can be significantly inhibited. CD4 + CD25 + Treg can regulate the immune status of the recipient through this effect on B cells so as to control the rejection , The role of induced immune tolerance.
