目的 建立一种可同时测定微量谷氨酸 (Glu) ,天门冬氨酸 (Asp) ,牛磺酸 (Tau) ,γ-氨基丁酸 (GABA) 4种氨基酸类神经递质的 HPL C分析方法 .方法 制备大鼠海马脑片 ,灌流取样 ,冻干再溶后 ,采用邻苯二甲醛 (OPA)柱前衍生 ,反相梯度洗脱的高效液相 /荧光检测法 ,测定其中 4种微量氨基酸的含量 .结果 各氨基酸的浓度与峰面积均呈现良好的线性关系 ,相关系数 (r)分别为 :Asp0 .992 0 ,Glu0 .96 6 7,Tau 0 .980 3和 GABA 0 .95 84 ;日内变异系数 (% )为 Glu 4 .3,Asp 4 .6 ,Tau6 .7和 GABA7.1;最低检出限 (pmol· L- 1 )为 :Asp 0 .17,Glu 0 .35 ,Tau 0 .6 8和 GABA 1.12 ,标准加入回收率 (% )分别为 :Asp 10 6± 7,Glu 10 1± 5 ,Tau 94± 6和GABA92± 11.结论 该法具有衍生化反应操作简便 ,流动相成本低 ,灵敏度高 ,特异性强 ,分析时间短的优点 ,而且同样适用于其他生物样品的测定

The four kinds of trace amino acids were determined by OPA pre column derivatization and reversed phase gradient elution. .Results The concentration of each amino acid showed a good linear relationship with the peak area, the correlation coefficients (r) were as follows: Asp0 .992 0, Glu0. 96 6 7, Tau 0 .980 3 and GABA 0 .95 84; The coefficient of variation (%) was Glu 4 .3, Asp 4 .6, Tau6 .7 and GABA7.1; the lowest detection limit (pmol · L-1) was: Asp 0 .17, Glu 0 .35, Tau 0. 6, and GABA 1.12 respectively, the standard recoveries (%) were as follows: Asp 10 6 ± 7, Glu 10 1 ± 5, Tau 94 ± 6 and GABA 92 ± 11. Conclusions The method has the advantages of simple derivatization, Low, high sensitivity, specificity, short analysis time, but also for the determination of other biological samples
