干扰素-γ、一氧化氮与血管平滑肌细胞增殖的关系

目的:研究干扰素γ与一氧化氮合酶、一氧化氮及血管平滑肌细胞增殖之间的关系。方法:将不同质量浓度的干扰素γ(250~1000U·ml-1)作用于培养的血管平滑肌细胞中,采用[3H]TdR掺入、细胞生长曲线和定量RTPCR等方法检测血管平滑肌细胞一氧化氮合酶基因表达,一氧化氮产生及血管平滑肌细胞增殖。结果:干扰素γ可以诱导血管平滑肌细胞表达一氧化氮合酶基因及酶蛋白,产生一氧化氮并呈质量浓度依赖。同时还发现干扰素γ抑制血管平滑肌细胞的DNA合成及细胞增殖与一氧化氮的产生是相平行的。结论:干扰素γ能通过诱导血管平滑肌细胞表达一氧化氮合酶基因产生一氧化氮来抑制血管平滑肌细胞增殖 Objective: To study the relationship between interferon  γ and nitric oxide synthase, nitric oxide and the proliferation of vascular smooth muscle cells. Methods: Interferon  γ (250 ~ 1000U · ml-1) with different concentrations was applied to cultured vascular smooth muscle cells. [3H] TdR incorporation, cell growth curve and quantitative RTPCR were used to detect blood vessels Nitric Oxide Synthase Gene Expression, Nitric Oxide Production and Vascular Smooth Muscle Cell Proliferation in Smooth Muscle Cells. Results: Interferon  γ can induce the expression of nitric oxide synthase gene and enzyme protein in vascular smooth muscle cells, producing nitric oxide in a concentration - dependent manner. Also found that interferon  γ inhibition of vascular smooth muscle cell DNA synthesis and cell proliferation and nitric oxide production are parallel. Conclusion: Interferon  γ can inhibit the proliferation of vascular smooth muscle cells by inducing nitric oxide production by nitric oxide synthase gene in vascular smooth muscle cells
