目的探讨端粒酶hTERT,p16及Topo-Ⅱa在星形细胞瘤组织中的表达及其相关性。方法应用组织芯片技术,S-P免疫组织化学方法检测64例不同级别的星形细胞瘤及10例正常脑组织中3种蛋白的表达情况。结果端粒酶hTERT,Topo-Ⅱa的蛋白表达率分别为50.0%与59.4%,随肿瘤级别的增高而增高,差异有显著性(P<0.05)。而p16在Ⅲ,Ⅳ级中蛋白表达明显低于Ⅰ,Ⅱ级星形细胞瘤及正常脑组织,且差异有显著性(P<0.05)。端粒酶hTERT蛋白表达与星形细胞瘤组织学分级、Topo-Ⅱa蛋白表达呈正相关。结论hTERT,p16及Topo-Ⅱa蛋白表达失调参与了星形细胞瘤的发生、发展,并影响星形细胞瘤的生物学行为。
Objective To investigate the expression and correlation of telomerase hTERT, p16 and Topo-Ⅱa in astrocytoma. Methods Tissue microarray technique and S-P immunohistochemistry were used to detect the expression of three proteins in 64 different grades of astrocytomas and 10 normal brain tissues. Results The protein expression rates of telomerase hTERT and Topo-Ⅱa were 50.0% and 59.4% respectively, which increased with the tumor grade increasing (P <0.05). However, the protein expression of p16 in grade Ⅲ and grade Ⅳ was significantly lower than those in grade Ⅰ and grade Ⅱ astrocytomas and normal brain tissues (P <0.05). Telomerase hTERT protein expression and histological grade of astrocytoma, Topo-Ⅱa protein expression was positively correlated. Conclusion The imbalanced expression of hTERT, p16 and Topo-Ⅱa is involved in the development of astrocytomas and the biological behavior of astrocytomas.
