目的 :研究卵巢癌血管内皮生长因子 (VEGF) m RNA异构体表达特征 ,探讨何种异构体在卵巢癌的发生发展中发挥优势作用。方法 :采用 RT- PCR、套式 PCR和测序分析对 30例卵巢癌组织 VEGF m RNA异构体表达进行相对定量检测。结果 :卵巢癌组织和卵巢正常组织表达 VEGF12 1、VEGF14 5、VEGF165、VEGF189m RNA,VEGF12 1表达水平显著高于其它 3种异构体 (P均 <0 .0 0 1)。 4种异构体在癌组织的表达水平均显著高于正常卵巢组织 (P均 <0 .0 5 )。结论 :卵巢癌组织高表达 VEGF12 1、VEGF14 5、VEGF165、VEGF189m RNA,以 VEGF12 1为主 ,提示 4种异构体均参与卵巢癌发生发展过程 ,VEGF12 1可能在其中发挥优势作用
OBJECTIVE: To study the expression of vascular endothelial growth factor (VEGF) m RNA isoforms in ovarian cancer and to explore which isoforms play an important role in the development of ovarian cancer. Methods: RT-PCR, nested PCR and sequencing analysis were used to detect the expression of VEGF m RNA isoforms in 30 cases of ovarian cancer. Results: The expression of VEGF12 1, VEGF14 5, VEGF165, VEGF189m RNA and VEGF12 1 in ovarian cancer tissues and normal ovarian tissues were significantly higher than those in the other three isoforms (all P <0.01). The expression of four isoforms in cancerous tissues were significantly higher than that in normal ovarian tissues (all P <0.05). CONCLUSION: VEGF12 1, VEGF14 5, VEGF165 and VEGF189mRNA are highly expressed in ovarian cancer tissues, mainly VEGF12 1, suggesting that all four isoforms are involved in the development and progression of ovarian cancer. VEGF12 1 may play a dominant role in this process
