目的:观察血塞通软胶囊对大鼠缺血心肌血管新生作用,并探讨其机制。方法:结扎大鼠左冠状动脉后,随机分为血塞通软胶囊组、麝香保心丸组、模型组,并设假手术组。6周后检测各组大鼠缺血心肌中微血管数(MVC)、微血管密度(MVD)及血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)、血小板衍生生长因子β(PDGF-β)、胰岛素样生长因子(IGF-1)蛋白的表达及其灰度值。结果:模型组、麝香保心丸组及血塞通软胶囊组大鼠心肌梗死(MI)边缘区MVC和MVD较假手术组明显增多(P<0.05);血塞通软胶囊组明显高于模型组(P<0.05),与麝香保心丸组相似(P>0.05)。模型组VEGF、bFGF、PDGF-β、IGF-1蛋白表达及其灰度值明显高于假手术组(P<0.05);血塞通软胶囊组明显高于模型组(P< 0.05),低于麝香保心丸组(P>0.05)。结论:血塞通软胶囊可促进大鼠缺血心肌血管新生,其机制与促进VEGF、bFGF、PDGF-β、IGF-1的表达有关。
Objective: To observe the effect of Xuesaitong soft capsule on the angiogenesis of ischemic myocardium in rats and to explore its mechanism. METHODS: After ligation of the left coronary artery in rats, they were randomly divided into Xuesaitong soft capsule group, Huxiang Baoxin Pill group and model group, and a sham operation group was set up. After 6 weeks, the number of microvessels (MVC), microvessel density (MVD) and vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF), platelet-derived growth factor beta (PDGF) in ischemic myocardium of each group were detected. -β), Insulin-like growth factor (IGF-1) protein expression and its gray value. RESULTS: Compared with the sham operation group, MVC and MVD in the marginal area of myocardial infarction (MI) in the model group, Huxiang Baoxin Pill group and Xuesaitong soft capsule group were significantly higher than those in the sham operation group (P<0.05); Higher than the model group (P<0.05), similar to the Huxiang Baoxin Pill group (P>0.05). The expression and gray value of VEGF, bFGF, PDGF-β and IGF-1 protein in the model group were significantly higher than those in the sham operation group (P<0.05). The Xuesaitong soft capsule group was significantly higher than the model group (P<0. 05), lower than Huoxiangxinxin Pills group (P>0.05). Conclusion: Xuesaitong soft capsule can promote angiogenesis of ischemic myocardium in rats, and its mechanism is related to the expression of VEGF, bFGF, PDGF-β and IGF-1.
