目的:评价吉西他滨对荧光素钾在携带荧光素酶报告基因的乳腺癌细胞株4T1luc胞内的外排动力学的影响。方法:以4T1luc细胞为模型,经不同浓度吉西他滨干预24 h后加入50μg·m L﹣1荧光素钾,应用生物发光成像技术连续动态检测各处理组生物发光信号强度随时间变化过程,计算荧光素钾的外排动力学参数。结果:荧光素钾浓度与生物发光信号光子量呈正相关。随着吉西他滨处理浓度增加,K逐渐增加,AUC逐渐降低(P<0.05)。结论:吉西他滨能够促进乳腺癌细胞胞内荧光素钾的外排动力学过程。
Objective: To evaluate the effect of gemcitabine on the efflux dynamics of fluorescein potassium in 4T1luc cells carrying luciferase reporter gene. Methods: With 4T1luc cells as model, 50μg · m L-1 fluorescein potassium was added after 24 h intervention with different concentrations of gemcitabine. The bioluminescence imaging technique was used to detect the dynamic changes of bioluminescence signal intensity in different treatment groups. Fluorescein Potassium efflux kinetic parameters. Results: There was a positive correlation between the concentration of fluorescein and photon of bioluminescence signal. With the increase of gemcitabine concentration, K gradually increased and AUC gradually decreased (P <0.05). Conclusion: Gemcitabine can promote the efflux kinetics of intracellular fluorescein potassium in breast cancer cells.
