目的定性研究视网膜缺血-再灌注过程中细胞死亡的方式。方法应用大鼠视神经结扎方法制成视网膜缺血-再灌注的动物模型,缺血时间60 min,再灌注3~72 h,应用透射电镜观察视网膜内层细胞的超微结构的改变。结果正常对照组没有发现视网膜细胞的异常改变。在缺血和再灌注的不同时相可以观察到的视网膜内层细胞发生两种不同模式的细胞死亡———进行性的细胞核和细胞的溶解和进行性的细胞和细胞核的浓缩。结论视网膜缺血再灌注过程中细胞死亡方式符合坏死和凋亡的特点,并持续存在于急性缺血损伤后的再灌注阶段,继续危害其他仍存活着的细胞。
Objective To qualitatively study the way of cell death during retinal ischemia-reperfusion. Methods Retinal ischemia-reperfusion model was established by ligating the optic nerve of rats. The ischemic time was 60 min and the reperfusion time was 3 to 72 h. The ultrastructure of retina was observed by transmission electron microscopy. Results No abnormal changes of retinal cells were found in normal control group. Two different patterns of cell death occurred in retinal inner cells observed at different phases of ischemia and reperfusion - progressive nuclear and cellular lysis and progressive cellular and nuclear condensation. Conclusion The pattern of cell death during retinal ischemia-reperfusion is consistent with the characteristics of necrosis and apoptosis. It persists in the reperfusion stage after acute ischemic injury and continues to harm other surviving cells.
