目的 探讨阿霉素 (ADM)与人转铁蛋白 (Tf)结合物对耐阿霉素 K5 6 2细胞株的体外杀伤作用。 方法 采用葡聚糖 (DEX) T- 10作为中介体、联接 Tf与 ADM制备结合物 Tf- ADM,用 MTT法比较结合物对耐ADM及 ADM敏感的 K5 6 2细胞的体外杀伤作用强度。 结果 结合物对耐 ADM的 K 5 6 2细胞株 72 h培养、MTT法测定结合物对耐 ADM的 K5 6 2细胞 IC5 0 为 4.9μmol,游离 ADM的 IC5 0 >17.2 4μmol,Tf对这两种细胞都没有体外杀伤作用。 结论 结合物 Tf- ADM可提高 ADM对耐 ADM的 K5 6 2细胞株的杀伤作用 ,此作用与 Tf无明显的关系。
Objective To investigate the killing effect of adriamycin (ADM) and human transferrin (Tf) on doxorubicin-resistant K562 cells in vitro. Methods Dextran (DEX) T-10 was used as the mediator, Tf-ADM was prepared by the combination of Tf and ADM. The anti-ADM and ADM-resistant K5 6 2 cells were compared in vitro by MTT assay. Results The conjugates were cultured for 72 h in ADM-resistant K562 cells. The IC50 values of the conjugates against ADM-resistant K562 cells were 4.9μmol and IC50> 17.2 4μmol of free ADM were determined by MTT assay. No cell killing effect in vitro. Conclusion The combination of Tf-ADM and ADM can increase the killing effect of ADM on K562 cell line resistant to ADM. This effect has no obvious relationship with Tf.
