目的:观察益气化痰方对慢性阻塞性肺疾病小鼠的肺功能及肿瘤坏死因子-α(TNF-α)、白细胞介素-17(IL-17)的影响。方法:将60只C57BL小鼠随机分成6组,分别是正常对照组、COPD模型组、低剂量中药组、中剂量中药组、高剂量中药组、地塞米松组,每组10只。香烟烟熏方法建立慢性阻塞性肺疾病小鼠模型,给药后分别观察小鼠肺功能以及对肺泡灌洗液中TNF-α、IL-17的变化。结果:与正常对照组相比,COPD模型组吸气峰流量(PIF)、呼气峰流量(PEF)及每分钟通气量(MV)数值均降低,差异均有统计学意义(P<0.01),低、中、高剂量的益气化痰方均能明显改善COPD小鼠气道的高反应状态,差异均有统计学意义(P<0.01,P<0.05)。与COPD模型组比较,中、高剂量组肺泡灌洗液中IL-17和TNF-α逐渐降低,差异均有统计学意义(P<0.01)。结论:益气化痰方能明显改善COPD小鼠的气道高反应,同时降低气道炎症反应。
Objective: To observe the effects of Yiqi Huatan Fang on pulmonary function, TNF-α and IL-17 in chronic obstructive pulmonary disease (COPD) mice. Methods: Sixty C57BL mice were randomly divided into 6 groups: normal control group, COPD model group, low dose traditional Chinese medicine group, middle dose traditional Chinese medicine group, high dose traditional Chinese medicine group and dexamethasone group. Cigarette smoked method to establish a mouse model of chronic obstructive pulmonary disease, after administration were observed in mice with lung function and changes of alveolar lavage fluid TNF-α, IL-17. Results: Compared with the normal control group, the values of PIF, PEF and MV in COPD model group were significantly lower than those in normal control group (P <0.01) The low, middle and high doses of Yiqi Huatan prescription can significantly improve the airway hyperresponsiveness in COPD mice, with statistical significance (P <0.01, P <0.05). Compared with the COPD model group, IL-17 and TNF-αin the bronchoalveolar lavage fluid of middle and high dose groups decreased gradually, the difference was statistically significant (P <0.01). Conclusion: Yiqi Huatan Decoction can significantly improve airway hyperresponsiveness in COPD mice and reduce airway inflammation.
