目的:研究安石榴苷对慢性支气管炎大鼠抗氧化能力的影响及其作用机制。方法:采用混合烟雾吸入法建立烟熏致大鼠慢性支气管炎模型,将大鼠随机分为:空白对照组、模型组、不同浓度安石榴苷(10、20及40mg/kg)处理组。处理结束后测定各组大鼠肺组织及血清中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性以及丙二醛(MDA)的含量。ELISA法检测各组大鼠血清中TNF-α、IL-8,IL-1β、IL-6的含量。Westernblot法检测PPARγ、PGC-1α、Nrf2、γ-GCS的表达。结果:模型组大鼠肺组织及血清中的SOD、CAT、GSHPx活性均显著低于对照组,MDA含量显著高于对照组。与模型组相比较,20、40mg/kg安石榴苷可显著增加大鼠肺组织及血清中的SOD、CAT、GSH-Px的活性,抑制MDA的产生。ELISA结果表明20、40mg/kg安石榴苷可显著抑制TNF-α、IL-8、IL-1β、IL-6的产生。Western blot结果表明安石榴苷可显著促进PPARγ、PGC-1α、Nrf2、γ-GCS的表达。结论:安石榴苷通过活化PPARγ/PGC-1α和Nrf2/γ-GCS信号通路,进而提高慢性支气管炎大鼠的抗氧化能力,缓解炎性反应。
Objective: To study the effect of Pomegranate on the antioxidant capacity of chronic bronchitis and its mechanism. Methods: The model of chronic bronchitis induced by smoked was established by mixed smoke inhalation method. The rats were randomly divided into control group, model group and different concentrations of punicrin (10, 20 and 40 mg / kg). After the treatment, the activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) Content. The levels of TNF-α, IL-8, IL-1β and IL-6 in serum of each group were detected by ELISA. The expression of PPARγ, PGC-1α, Nrf2 and γ-GCS were detected by Western blot. Results: The activities of SOD, CAT and GSHPx in the lung tissue and serum of the model group were significantly lower than those of the control group, and the MDA content was significantly higher than that of the control group. Compared with the model group, 20,40 mg / kg punicalagin significantly increased the activity of SOD, CAT and GSH-Px in lung tissue and serum, and inhibited the production of MDA. ELISA results showed that 20, 40mg / kg punicala could significantly inhibit the production of TNF-α, IL-8, IL-1β and IL-6. Western blot results showed that pomegranate can significantly promote the expression of PPARγ, PGC-1α, Nrf2, γ-GCS. Conclusion: Pomegranate can activate PPARγ / PGC-1α and Nrf2 / γ-GCS signal pathways to improve the anti-oxidative capacity and relieve the inflammatory reaction in rats with chronic bronchitis.
