目的:研究血府逐瘀汤含药血清对大鼠肾小球系膜细胞(RMC)经ox-LDL刺激后肿瘤坏死因子-α(TNF-α)和血凝素样氧化低密度脂蛋白受体-1(LOX-1)m RNA表达的影响,探讨血府逐瘀汤对RMC的保护性作用。方法:使用50μg/m L ox-LDL刺激RMC 24h,用包括不同浓度血府逐瘀汤、辛伐他汀含药血清进行干预24h,使用荧光定量PCR(RT-PCR)检测RMC中TNF-α和LOX-1的m RNA表达。结果:与模型组相比,不同剂量血府逐瘀汤含药血清和辛伐他汀含药血清均可不同程度地降低TNF-α及LOX-1的m RNA表达(P<0.01),血府逐瘀汤含药血清的干预作用随浓度升高而升高,高浓度血府逐瘀汤含药血清的干预作用较辛伐他汀含药血清强(P<0.01)。结论:血府逐瘀汤含药血清可减少RMC中TNF-α、LOX-1的表达,减轻ox-LDL对RMC的损伤作用。
OBJECTIVE: To investigate the effect of Xuefu Zhuyu Decoction-containing serum on the expression of tumor necrosis factor-α (TNF-α) and hemagglutinin-like oxidized low density lipoprotein (r-LDL) in rat glomerular mesangial cells (-1) (LOX-1) m RNA, and explore the protective effect of Xuefu Zhuyu Decoction on RMC. Methods: The RMCs were stimulated with ox-LDL (50μg / mL) for 24 h. Serum containing Xuefu Zhuyu Decoction and Simvastatin at different concentrations were used for 24h intervention. The levels of TNF-α and M RNA expression of LOX-1. Results: Compared with model group, serums of Xuefuzhuyu Decoction and simvastatin containing different doses could reduce the mRNA expression of TNF-α and LOX-1 to varying degrees (P <0.01) The intervention effect of Zhuyu Decoction containing serum increased with the increasing of concentration, and the effect of Xuefu Zhuyu Decoction containing high concentration was stronger than that of simvastatin containing serum (P <0.01). Conclusion: Xuefu Zhuyu Decoction-containing serum can reduce the expression of TNF-α and LOX-1 in RMC and reduce the injury of RMC induced by ox-LDL.
