目的:观察抑郁症睡眠障碍(SDD)大鼠自主行为及血清TT3(三碘甲状腺原氨酸),TT4(甲状腺素),FT3(游离三碘甲状腺原氨酸),FT4(游离甲状腺素),TSH(促甲状腺素)表达变化,探讨甲状腺激素水平与抑郁症睡眠障碍的关联及逍遥散的干预作用。方法:48只Wistar大鼠进行敞箱行为(OFT)试验,根据得分将大鼠随机分为4组,正常组、模型组、米氮平组、逍遥散组。除正常组外,其余各组共接受21 d不同的应激。造模开始当天应激前1 h模型组、米氮平组、逍遥散组分别给予等容量生理盐水、米氮平、逍遥散水提液。21 d计算行为得分后处死大鼠,取大鼠血清置-20℃冰箱中备测TT3,TT4,FT3,FT4,TSH水平。结果:21 d后模型组自主活动较应激前显著减少(P<0.01),与模型组比较,逍遥散组及米氮平组大鼠的自主活动明显增加(P<0.01),2组与正常组无差异;各组血清TT3,TT4水平变化无差异;模型组FT3水平下降,FT4水平上升,TSH水平无变化,米氮平组、逍遥散组与模型组比较,无统计学意义。结论:逍遥散具有缓解抑郁症睡眠障碍的效应,但对甲状腺激素水平变化无影响。
OBJECTIVE: To observe the autonomic behavior and serum TT3 (TTX), TT4 (thyroxine), FT3 (free triiodothyronine), FT4 (free thyroxine) in SDD rats, TSH (thyroid-stimulating hormone) expression changes to explore the relationship between thyroid hormone levels and sleep disorders in depression and Xiaoyaosan intervention. Methods: Forty-eight Wistar rats were subjected to the open-box behavior (OFT) test. According to the scores, the rats were randomly divided into four groups: normal group, model group, mirtazapine group and Xiaoyao powder group. In addition to the normal group, the remaining groups received a total of 21 d different stress. The model group, mirtazapine group and Xiaoyao powder group were given equal volumes of normal saline, mirtazapine and Xiaoyaosan water extract respectively 1 h before the start of modeling. The rats were sacrificed after 21 days of behavioral score calculation, TT3, TT4, FT3, FT4 and TSH levels were prepared in the refrigerator at -20 ℃. Results: Compared with the model group, the autonomic activities of model group increased significantly (P <0.01) after 21 days, compared with the model group (P <0.01) The levels of TT3 and TT4 in serum of each group were not significantly different. The levels of FT3, FT4 and TSH in the model group were not changed. There was no significant difference between the mirtazapine and Xiaoyaosan groups and the model group. Conclusion: Xiaoyaosan has the effect of alleviating sleep disorders in depression, but it has no effect on the level of thyroid hormone.
