目的:探讨左旋咪唑(LMS)对实验性自身免疫性脑脊髓炎(EAE)脊髓IFN--γ、TGF-β1mRNA表达的影响。方法:用GPSCH免疫Wistar大鼠制作EAE模型,分别在免疫前(LMS1组)、免疫同时(LMS2组)、免疫后(LMS3组)给LMS10mg·kg-1。比较各组行为学和组织学的变化,用RT-PCR测脊髓IFN-和TGF-β1mRNA的表达。结果:LMS1组和LMS2组的行为学表现和炎细胞浸润加重(P<0.01,P<0.05)。LMS3组出现EAE复发。与EAE组比,LMS1组和LMS2组脊髓IFN-mRNA表达升高,TGF-β1mRNA表达降低,但仅LMS2组有统计学意义(P<0.05)。结论:LMS可升高脊髓IFN-mRNA表达,降低TGF-β1mRNA表达,提示LMS对EAE的促发作用可能与Th1/Th2失衡有关。
Objective: To investigate the effect of levamisole on the expression of IFN - γ and TGF - β1 mRNA in spinal cord of experimental autoimmune encephalomyelitis (EAE). Methods: Wistar rats were immunized with GPSCH to make EAE model. The rats were killed before LMS1 immunization (LMS1 group) and immunized with LMS2 group (LMS3 group). The behavioral and histological changes of each group were compared. The mRNA expression of IFN- and TGF-β1 in the spinal cord was measured by RT-PCR. Results: The behavioral findings and infiltration of inflammatory cells in LMS1 and LMS2 groups were aggravated (P <0.01, P <0.05). EAE recurrence occurred in LMS3 group. Compared with EAE group, the expression of IFN-γ mRNA and the mRNA of TGF-β1 in LMS1 group and LMS2 group were lower than that in EAE group, but only in LMS2 group and LMS2 group (P <0.05). Conclusion: LMS can increase the expression of IFN-γ mRNA in the spinal cord and decrease the expression of TGF-β1 mRNA, suggesting that LMS may contribute to the imbalance of Th1 / Th2.
