目的:探讨大黄、虫对药对雌、孕激素负荷大鼠子宫肌瘤模型雌激素受体亚型ERα、ERβ的影响及作用机理。方法:70只SD雌性大鼠随机分成正常对照组、模型组、大黄虫小剂量组、大黄虫中剂量组、大黄虫大剂量组、大黄虫丸组、桂枝茯苓胶囊组。采用雌、孕激素负荷法建立大鼠子宫肌瘤模型,用SABC免疫组化法测定大鼠子宫平滑肌细胞ERα、ERβ的表达水平。结果:模型大鼠子宫平滑肌细胞ERα、ERβ的表达显著增高(P<0.01),各治疗组的ERα、ERβ较模型组均下降(P<0.01)。结论:大黄、虫对药可抑制子宫肌瘤平滑肌细胞ERα、ERβ的表达,这可能是该药治疗子宫肌瘤的主要机理之一。
Objective: To investigate the effects of rhubarb and pinworm on estrogen and progesterone-loaded estrogen receptor subtype ERα and ERβ in uterine leiomyoma model rats and its mechanism. Methods: Seventy SD female rats were randomly divided into normal control group, model group, small dose group of Rhubarb, large dose group of rhubarb, large dose group of rhubarb, group of rhubarb and wormwood. Uterine fibroids model was established by estrogen and progesterone loading method. The expression of ERα and ERβ in rat uterine smooth muscle cells was detected by SABC immunohistochemistry. Results: The expression of ERα and ERβ in uterine smooth muscle cells of model rats were significantly increased (P <0.01). The levels of ERα and ERβ in each treatment group were lower than those in model group (P <0.01). Conclusion: Rhubarb and pinworm can inhibit the expression of ERα and ERβ in uterine fibroids smooth muscle cells, which may be one of the main mechanisms of the treatment of uterine fibroids.
