目的:建立甘肃铨水大黄药材HPLC指纹图谱分析方法,为铨水大黄的质量控制提供依据。方法:用Cosmosil 5C18-PAQ(250 mm×4.6 mm,5μm)色谱柱,以乙腈-0.1%乙酸水溶液为流动相进行梯度洗脱,流速:1 mL/min,柱温:30℃,检测波长:280 nm;测定收集的21份大黄样品,对数据进行相似度评价、主成分分析和聚类分析。结果:建立了铨水大黄HPLC指纹图谱分析方法;根据相似度评价、主成分分析和聚类分析结果,筛选10份样品建立了铨水大黄的特征指纹图谱。结论:在建立的HPLC条件下各成分分离较好,可用于铨水大黄的分析和质量控制。
OBJECTIVE: To establish a method for the HPLC fingerprint analysis of rhubarb in Quanshuanshui, Gansu Province, and provide the basis for the quality control of rhubarb. METHODS: The elution was performed on a Cosmosil 5C18-PAQ column (250 mm × 4.6 mm, 5 μm) with acetonitrile-0.1% aqueous acetic acid as mobile phase at a flow rate of 1 mL / 280 nm; 21 rhubarb samples were collected and analyzed for similarity, principal component analysis and cluster analysis. Results: The HPLC fingerprinting method of Rhubarb was established. According to the similarity evaluation, principal component analysis and clustering analysis results, 10 samples were selected to establish the characteristic fingerprints of Rheum. Conclusion: The separation of each component under the established HPLC conditions is good, which can be used for the analysis and quality control of Rheum.
