目的:研究凉血活血方对腹腔感染脓毒症大鼠肠组织纤溶及胶原代谢的影响,探讨其作用机制。方法:Wistar大鼠50只,雌雄各半,随机分为假手术组、模型组、凉血活血方治疗组。模型组大鼠以丝线结扎盲肠末端,针头穿孔,挤出少许粪便后关腹,制作腹腔感染脓毒症模型。治疗组于术后开始给予活血凉血方进行ig治疗,每日1次,连续治疗72 h,假手术组和模型组给予等体积蒸馏水ig。统计动物死亡率,ELISA法测定存活大鼠肠组织的组织纤溶酶原激活物(t-PA),Ⅰ型纤溶酶原激活物抑制因子(PAI-1),结缔组织生长因子(CTGF),成纤维细胞生长因子-10(FGF-10),Ⅰ型胶原,Ⅲ型胶原含量或活性。结果:凉血活血方治疗组动物死亡率为25%,与模型组(65%)比较显著降低(P<0.05);与假手术组相比,模型组t-PA水平显著降低(P<0.01),而PAI-1,CTGF,FGF-10,I型胶原,III型胶原显著升高(P<0.01);与模型组相比,凉血活血方治疗组t-PA水平显著回升(P<0.01),而PAI-1,CTGF,FGF-10,Ⅰ型胶原,Ⅲ型胶原水平显著降低(P<0.01)。结论:凉血活血方具有良好的纤溶调节、抗胶原增生作用,对腹腔感染脓毒症具有良好的治疗作用。
Objective: To study the effect of Liangxue Huoxue Recipe on fibrinolytic and collagen metabolism in intestinal tissue of rats with sepsis induced by intraperitoneal infection and to explore its mechanism. Methods: Fifty Wistar rats were randomly divided into sham operation group, model group and Liang Xue Huoxue decoction group. The rats in the model group were ligated with the ends of the cecum via silk thread and perfused the needles. After excreting a little feces, they were closed and abdominal cavity was made. The model of sepsis was made by intraperitoneal injection. The treatment group was given ig treatment with Huoxue Liangxue Recipe after operation, once a day for 72 hours. The sham operation group and model group were given equal volume of distilled water ig. The mortality of the animals was measured. Tissue plasminogen activator (t-PA), type I plasminogen activator inhibitor (PAI-1) and connective tissue growth factor (CTGF) , Fibroblast growth factor-10 (FGF-10), type I collagen, type III collagen content or activity. Results: Compared with sham-operated group, the t-PA level in the model group was significantly lower than that in the sham-operated group (P <0.01), and the death rate of the model group was significantly lower than that of the model group (65% ), While PAI-1, CTGF, FGF-10, type I collagen and type III collagen were significantly increased (P <0.01). Compared with the model group, t- 0.01). However, the levels of PAI-1, CTGF, FGF-10, collagen I and collagen Ⅲ were significantly decreased (P <0.01). Conclusion: Liangxuehuoxue Fang has good fibrinolytic regulation and anti-collagen hyperplasia, and has a good therapeutic effect on sepsis caused by abdominal infection.
