目的探讨p5 3凋亡刺激蛋白基因(ASPP)的抑制蛋白iASPP在乳腺浸润性小叶癌中的表达及其临床意义。方法采用RT-PCR技术扩增乳腺浸润性小叶癌组织中iASPP mRNA;同时应用Quality One软件分析iASPP扩增产物的相对含量,分析iASPP-mRNA与临床病理因素之间的关系。结果 3 9例浸润性小叶癌组织中有3 4例表达iASPP(8 7.2%),5例不表达(1 2.8%);癌旁组织中均不表达iASPP。iASPP-mRNA在乳腺浸润性小叶癌中的表达明显增高,与癌旁组织相比差异具有显著性(P<0.0 1)。不同的年龄组iASPP-mRNA的表达差异也有显著性;TNMⅢ、Ⅳ期乳腺癌组织中iASPP-mRNA的表达显著高于Ⅰ、Ⅱ期患者(P<0.0 1);有淋巴结转移的癌组织中iASPP-mRNA的表达也高于无淋巴结转移者;iASPP-mRNA的表达与雌、孕激素受体的表达无关。结论 iASPP-mRNA在乳腺浸润性小叶癌中高表达。检测iASPP可以为乳腺癌的诊断、个体化治疗及预后提供参考。
Objective To investigate the expression and clinical significance of iASPP, an inhibitor of p5 3 apoptosis-stimulatory protein gene (ASPP), in invasive lobular breast cancer. Methods The iASPP mRNA in breast invasive lobular carcinoma was amplified by RT-PCR. The relative content of iASPP amplification products was analyzed by Quality One software. The relationship between iASPP-mRNA and clinical pathological factors was analyzed. RESULTS: Thirty-three infiltrating lobular carcinoma tissues showed iASPP (8 7.2%) in 34 cases and no expression in 5 cases (14.8%). No paracancerous tissues expressed iASPP. The expression of iASPP-mRNA in breast invasive lobular carcinoma was significantly higher than that in paracancerous tissues (P <0.01). The expression of iASPP-mRNA in different age groups was also significantly different. The expression of iASPP-mRNA in TNMⅢ and Ⅳ breast cancer tissues was significantly higher than that in stage Ⅰ and Ⅱ patients (P <0.01); iASPP -mRNA was also higher than those without lymph node metastasis; iASPP-mRNA expression and estrogen and progesterone receptor expression has nothing to do. Conclusions iASPP-mRNA is highly expressed in invasive lobular breast tumors. Detection of iASPP can provide a reference for the diagnosis of breast cancer, individualized treatment and prognosis.
