目的:在已证明条件免疫反应(CIR)能够有效治疗大鼠胶原性关节炎(CIA)的前期研究基础上,进一步探讨其对大鼠CIA的作用机制。方法:建立大鼠CIA模型。将模型大鼠分为5组:CIR组,以樟脑气味为条件刺激,甲氨喋呤(MTX) +泼尼松(Pred)为非条件刺激,两者结合7次( 7天)后可建立条件免疫反应,然后每日再现条件刺激,条件刺激与非条件刺激每周结合1次,共4周;MTX +Pred组,MTX +Pred治疗4周;MTX +Pred减量组,MTX +Pred治疗7天内每天1次,7天后每周1次,共4周;单纯闻樟脑气味组,单纯闻樟脑气味4周;空白对照组,安慰剂治疗4周。用流式细胞技术检测大鼠外周血CD4 +T淋巴细胞活化(表达CD71)及表达烟碱样乙酰胆碱受体α7亚单位(nAChRα7)和胆碱乙酰转移酶(ChAT)的状态;免疫组织化学检测nAChRα7、ChAT、CD6 8(巨噬细胞标志)和TNFα在关节滑膜细胞的表达。结果:治疗1周后CIR组大鼠外周血CD4 +T淋巴细胞表达nAChRα7和ChAT明显高于其他各组(P <0 0 1) ;CIR组与MTX +Pred组大鼠,2周后外周血活化的CD4 +T淋巴细胞(表达CD71)明显减少(P <0 0 1) ,3周后大鼠关节炎指数明显低于其他各组(P <0 0 1) ;4周后滑膜组织表达CD6 8及TNFα明显降低(P <0 0 5 ) ;CIR组大鼠表达nAChRα7和ChAT明显高于其他各组(P <0 0 1)。结论:以樟脑气味为
OBJECTIVE: To further investigate its mechanism of action on rat CIA, based on the previous studies that the conditional immune response (CIR) can effectively treat rat collagen-induced arthritis (CIA). Methods: The rat CIA model was established. The model rats were divided into 5 groups: CIR group, stimulated by camphor odor, methotrexate (MTX) and prednisone (Pred) as unconditioned stimuli. The combination of the two can be established after 7 times MTX + Pred group, MTX + Pred for 4 weeks; MTX + Pred reduction group, MTX + Pred treatment group 7 days once a day, once a week for 7 days after a total of 4 weeks; simple camphor smell group, smell camphor smell for 4 weeks; blank control group, placebo for 4 weeks.
