目的:研究丹参及大黄对急性坏死性胰腺炎(ANP)早期肺损伤肺组织中性粒细胞凋亡及MMP-2、9表达的影响。方法:将60只大鼠随机分为4组,即正常组,模型组,丹参、大黄治疗组。采用逆行注射法建立大鼠SAP模型,观察检测外周血中性粒细胞凋亡,肺组织的中性粒细胞凋亡,MMP-2,9的表达和组织学变化及丹参、大黄对其影响。结果:模型组中性粒细胞凋亡延迟及肺组织MMP-2,9明显高于正常组和丹参大黄治疗组。丹参、大黄组的肺组织MMP-2,9表达明显降低,组织通透性降低。结论:SAP早期的肺损伤伴有组织中性粒细胞凋亡延迟及MMP-2,9的过度表达;丹参、大黄可降低组织MMP-9的表达,减轻肺组织损伤。
Objective: To study the effect of salvia miltiorrhiza and rhubarb on the apoptosis of lung neutrophil and the expression of MMP-2, 9 in acute lung injury of acute necrotizing pancreatitis (ANP). Methods: Sixty rats were randomly divided into four groups: normal group, model group, Salvia miltiorrhiza and rhubarb treatment group. The rat model of SAP was established by retrograde injection. The apoptosis of neutrophils, the apoptosis of neutrophils in lung tissue, the expression of MMP-2, MMP-9, histological changes and the effects of Danshen and rhubarb on it were observed. Results: The delayed apoptosis of neutrophil and the expression of MMP-2,9 in lung tissue of model group were significantly higher than those of normal group and rhubarb-treated group. Salvia miltiorrhiza, rhubarb group of lung tissue MMP-2,9 expression was significantly reduced, tissue permeability decreased. CONCLUSIONS: Early lung injury in SAP is associated with delayed apoptosis of the neutrophils and overexpression of MMP-2 and MMP-9. Salviae miltiorrhizae and rhubarb can decrease the expression of MMP-9 and decrease the lung injury.
