通过细胞学和形态学观察,从(陆地棉×索马里棉)异源六倍体的陆地棉回交后代中,鉴定出2rrrrrrrrn个异源单体附加系,用 RARD 方法对其附加的染色体来源进行分子鉴定和区分。在 160 条引物中,筛rrrrrrrrn选出引物SBSG11 能特异标记异源单体附加系Ⅰ所附加的索马里棉染色体,特征带长约600 bp ;引物rrrrrrrrnSBSC03 能特异标记异源单体附加系Ⅱ所附加的索马里棉染色体,特征带长约700 bp;引物SBSE07 和rrrrrrrrnSBSE08 分别扩增附加第Ⅰ和附加系Ⅱ产生共有的特征带。高强纤维索马里棉异源附加系对棉花品种rrrrrrrrn改良可能具有重要的价值。
Through cytological and morphological observation, we identified 2 × r r r r r r r r n from backcrossing of upland cotton (Heterosigmoia x Somali cotton) A heterologous monomer addition line, with RARD method of its additional source of chromosome molecular identification and distinction. Of the 160 primers, SBSG11 was able to specifically label the additional Somalia cotton chromosome attached to the heterologous monomer attachment system Ⅰ, with a characteristic length of about 600 bp. The primer SBSC03 could specifically label the additional Somalia cotton chromosome attached to the heterologous monomer II, and the characteristic band was about 700 bp. The primers SBSE07 and SBSE08 amplifies the additional signature bands I and II, respectively, to generate common signature bands. Heterologous addition lines of high-fiber Somali cotton may have important value for the improvement of cotton varieties.
