目的研究补骨脂中9种黄酮类化合物构效关系及对成骨细胞的增殖、分化活性。方法采用从绝经后妇女股骨松质骨分离得到的成骨细胞,利用噻唑蓝法检测黄酮类化合物对成骨细胞增殖的影响,同时测量成骨细胞碱性磷酸酶(ALP)活性来评价黄酮类化合物对人成骨细胞分化的影响。结果 9种黄酮化合物中,补骨脂二氢黄酮药物组对成骨细胞增殖、分化的活性最强,通过对9中化合物的EC50值和各药物组成骨细胞酶活性比较,发现二氢黄酮母核、7位羟基、6位异戊二烯基的存在对成骨细胞的增殖、分化具有显著的作用。结论补骨脂中黄酮类化合物对成骨细胞的增殖、分化作用与其结构有一定的相关性。
Objective To study the structure-activity relationship and the activity of osteoblast proliferation and differentiation of nine flavonoids in psoralen. Methods Osteoblasts isolated from femoral cancellous bone of postmenopausal women were used to detect the effect of flavonoids on the proliferation of osteoblasts by using thiazolyl blue method. The activity of alkaline phosphatase (ALP) in osteoblasts was measured to evaluate the effect of flavonoids Effects of Compounds on Human Osteoblast Differentiation. Results Among the nine flavonoids, psoralen group had the strongest activity on proliferation and differentiation of osteoblasts. Comparing the EC50 values of nine compounds with the activity of each group, The existence of nucleus, 7-hydroxyl group and 6-position isoprenyl group have a significant effect on the proliferation and differentiation of osteoblasts. Conclusion The flavonoids in psoralen have some correlations with the proliferation and differentiation of osteoblasts.
