目的 了解TT病毒 (TTV)的组织嗜性。方法 从 5只实验感染病毒的恒河猴各组织中抽提DNA ,分别用病毒双链探针或单链负义探针 ,与吸附在纤维膜上的组织DNA进行斑点杂交。结果 双链探针与肝、骨髓、脾、胃、小肠、血清和大肠杂交阳性 ,表明这些组织中都有TT病毒的存在。单链负义探针与各组织杂交 ,仅肝、骨髓和小肠为阳性 ,提示可能存在作为病毒复制中间体的正义单链DNA。结论 肝、骨髓和小肠可能是病毒的复制场所 ,故TT病毒可能具嗜肝性 ,嗜小肠性和嗜骨髓性
Objective To understand the tissue tropism of the TTV virus. Methods DNA was extracted from five experimental rhesus monkeys infected with virus. Dot double-stranded hybridization was performed with DNA double-stranded probe or single-stranded negative probe, respectively, on the tissue DNA adsorbed on the fiber membrane. Results Double-stranded probes were positive for hybridization to liver, bone marrow, spleen, stomach, small intestine, serum and large intestine, indicating the presence of TT virus in these tissues. The single-stranded negative probe hybridized with each tissue and positive only in the liver, bone marrow and small intestine, suggesting that there might be sense single-stranded DNA as a viral replication intermediate. Conclusion The liver, bone marrow and small intestine may be the site of replication of the virus, so the TT virus may have hepatomegaly, minor intestinal and myeloablative
