目的:考察西洋参冠瘿组织培养条件及其对多糖含量的影响。方法:采用正交设计法优化考察了转基因西洋参冠瘿组织的培养条件(如培养基、接种量、培养基pH值和无机元素等)及其对多糖含量的影响。结果:(1)该转基因组织能够在无激素的MS固体培养基中生长,并累积多糖;(2)得到了MS固体培养条件下冠瘿组织生长和多糖含量随时间变化的规律:培养21 d时,多糖含量最高,培养24 d时,冠瘿组织达到最大的生物量;(3)培养基pH值为5.6时,西洋参冠瘿组织生长状态良好,冠瘿组织生长量最高,多糖含量最高;(4)冠瘿组织接种量在4~7 g(新重每瓶)时,对西洋参冠瘿组织生长较有利,但接种量对多糖含量无明显影响。结论:此培养方法能实现西洋参冠瘿组织多糖的高累积。
OBJECTIVE: To investigate the tissue culture conditions and the effect on the content of polysaccharides in American ginseng. Methods: The orthogonal design method was used to optimize the culture conditions (such as medium, inoculum size, medium pH value and inorganic elements) of transgenic American Ginseng and its effect on polysaccharide content. Results: (1) The transgenic tissue could grow in hormone-free MS solid medium and accumulate polysaccharides; (2) The regularity of the growth and the content of polysaccharides of crown gall with time under MS solid culture conditions were obtained: (3) When the pH value of culture medium was 5.6, the growth of crown gynogenetic tissue of American ginseng was good, the growth of crown gizzard tissue was the highest, and the content of polysaccharide was the highest. (4) When the inoculum size of crown gall was 4 ~ 7 g (new weight per bottle), the growth of crown gall tissue of American ginseng was more favorable, but the inoculum size had no significant effect on polysaccharide content. Conclusion: This culture method can achieve high accumulation of polysaccharides in the crown ginseng tissue of American ginseng.
