目的探讨β_1肾上腺素能受体(β_1-AR)常见基因多态性对高血压病患者左心室肥厚的影响。方法高血压病患者114例分为合并左心室肥厚(EH-LVH组,50例)和不合并左心室肥厚者(EH组,64例)两组,对照组为同期正常体检者54例。采用聚合酶链反应-限制性片段长度多态性检测β_1-AR Gly389Arg和Ser49Gly位点的单核苷酸基因多态性。结果EH-LVH组患者β_1-AR Arg389Arg基因型比例高于EH组和对照组(86.0%vs.65.6%和51.9%)(P<0.05);EH-LVH组β_1-AR Arg389等位基因频率高于EH组和对照组(90.0%vs.78.1%和71.2%)(P<0.05)。三组β_1-AR Ser49Gly基因型和等位基因频率差异无统计学意义(P>0.05)。结论携带β_1-AR Arg389等位基因的高血压病患者更易出现左心室肥厚。
Objective To investigate the effect of common gene polymorphisms of β 1 adrenergic receptor (β 1 -AR) on left ventricular hypertrophy in patients with essential hypertension. Methods One hundred and fourteen patients with hypertension were divided into two groups: left ventricular hypertrophy (EH-LVH group, 50 cases) and left ventricular hypertrophy (EH group, 64 cases). The control group consisted of 54 normal subjects at the same period. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to detect single nucleotide polymorphisms in the Gly389Arg and Ser49Gly sites. Results The genotype frequency of β 1-AR Arg389Arg in EH-LVH group was significantly higher than that in EH group and control group (86.0% vs 65.6% and 51.9%, P <0.05) In the EH group and the control group (90.0% vs 78.1% and 71.2%, respectively) (P <0.05). There was no significant difference in genotype and allele frequency between the three groups of β1-AR Ser49Gly (P> 0.05). Conclusion Hypertension patients with β 1-AR Arg389 allele are more likely to have left ventricular hypertrophy.
