采用造血祖细胞体外培养。造血生长因子生物活性检测等实验血液学技术,研究维生素A缺乏(VAD)对大鼠红系早期祖细胞(BFU-E)增殖调控的影响。结果发现,与对照组比较,VAD使BFU-E集落增殖明显减少;用VAD的脾及骨髓基质细胞培养上清液明显抑制BFU-E集落增殖。结果提示,VAD可能通过抑制造血生长因子白细胞介素3(IL-3)和粒单系集落刺激因子(GM-CSF)等表达而影响红系造血。
Hematopoietic progenitor cells cultured in vitro. Hematopoietic growth factor bioassay and other experimental hematological techniques to study the effects of vitamin A deficiency (VAD) on the proliferation of rat erythroblast precursor progenitor cells (BFU-E). The results showed that, compared with the control group, VAD significantly reduced the proliferation of BFU-E colonies; the supernatant of VAD spleen and bone marrow stromal cells significantly inhibited the proliferation of BFU-E colonies. The results suggest that VAD may affect erythroid hematopoiesis by inhibiting the expression of hematopoietic growth factor interleukin 3 (IL-3) and granulocyte colony stimulating factor (GM-CSF).
