目的:建立玳玳总黄酮提取物主成分体内外正常与病理模型大鼠UPLC-MS特征图谱分析法,为玳玳总黄酮提取物入血成分及药效物质基础的阐明奠定基础。方法:采用UPLC-MS法,建立并比较玳玳总黄酮提取物体外特征图谱、正常和高脂血症模型大鼠灌胃玳玳总黄酮提取物后体内特征图谱。结果:经优选最佳色谱与质谱条件,并确立最佳采血时间及样品前处理方法后,质控9个色谱峰,相对保留时间RSD<0.5%,相对峰面积RSD<2.9%,提取回收率>75%,建立了玳玳总黄酮提取物体内外特征图谱模式。结论:所建立的特征图谱稳定、准确,可用于玳玳总黄酮提取物体外和正常及病理状态下的体内外成分分析评价,为“活性特征图谱”评价提供有益的探索。
OBJECTIVE: To establish a UPLC-MS characterization map of the main components of Tohto Daiwa flavonoids extract in vivo and in vitro, and to lay a foundation for elucidating the blood components and pharmacodynamic substances of Tohto Daiibei flavonoids extract. Methods: The UPLC-MS method was used to establish and compare the in vitro characteristics of total flavonoids from Toadia daidzein. In vivo and in vivo profiles of total flavonoids from Toadia daidzein were observed in normal and hyperlipidemic rats. Results: The optimum chromatographic and mass spectrometric conditions, and the establishment of the best sampling time and sample preparation methods, quality control 9 peaks, relative retention time RSD <0.5%, relative peak area RSD <2.9%, recovery rate > 75%, established the Toisai total flavonoids extract patterns in vitro and in vivo profiles. Conclusion: The established characteristic map is stable and accurate. It can be used to evaluate the in vitro and in vivo components of total flavonoids from TomatiWhitikis in vitro and in normal and pathological conditions, and to provide a useful exploration for the evaluation of “active characteristics map”.
