目的 观察牛初乳短链胰岛素样生长因子-1(BCtIGF-1)对糖尿病肾病的保护作用及探讨其可能的作用机制。方法采用生化法、放射免疫法、荧光分析法及逆转录.聚合酶链式反应(RT-PCR)技术等进行研究。结果(1)糖尿病治疗组的空腹血糖、甘油三酯、果糖胺明显低于糖尿病对照组;(2)糖尿病治疗组24h尿白蛋白排泄率、24h尿量、24h内生肌酐清除率低于糖尿病对照组;(3)电镜观察大鼠肾脏病理切片发现BCtIGF-1对糖尿病大鼠肾脏损害有改善作用;(4)糖尿病对照组肾组织IGF-1mRNA表达明显低于正常对照组,糖尿病对照组与糖尿病治疗组无明显异常;(5)糖尿病治疗组肾组织胶原含量及糖基化终末产物(AGEs)、尿血栓素B2/6-酮-前列环素1α(TXB2/6-keto-PGF1α)低于糖尿病对照组。结论(1)BCtIGF-1可以明显降低STZ糖尿病大鼠的血糖、血脂;(2) BCtIGF-1不会引起肾皮质 IGF-1mRNA表达的增加;(3) BCtIGF-1对 STZ糖尿病大鼠肾脏有保护作用,其保护机制是由于BCtIGF-1对血糖、血脂的改善;此外,可能与其减少紧组织胶原及AGEs含量,改善前列腺素-血栓素合成的不平衡?
Objective To observe the protective effect of bovine colostrum short chain insulin-like growth factor-1 (BCtIGF-1) on diabetic nephropathy and to explore its possible mechanism. Methods Biochemical methods, radioimmunoassay, fluorescence analysis and reverse transcription. Polymerase chain reaction (RT-PCR) technology to study. Results (1) The fasting blood glucose, triglyceride and fructosamine in the diabetic group were significantly lower than those in the diabetic control group.
